I don't get the "Usage" part.

does it mean to to to use the commands below?

And when I do run the command I get:

Cannot open assembly 'tuxlol.exe': No such file or directory

Here's what I did. Download the tar.gz and throw it onto your desktop. Right click and click extract here (or open it and extract it) 

A file named Release will appear on your desktop. Open it and right click somewhere in the folder and click on Open in Terminal. Now that the terminal destination is set to your folder enter the command it tells you. To make it a little quicker for you with PoL (instead of Wine) the command is

mono tuxlol.exe patch --dir "/home/USERNAME/.PlayOnLinux/wineprefix/LeagueOfLegends/drive_c/Riot Games/League of Legends/"

Fill in your system user where it says USERNAME.

Hit enter and it will start patching. Make sure League is up to date first though so you don't have to repatch in case it isn't updated.

You should have been doing that with the last script. When League updates some icons get reverted back. Not always all of them that were red before, but usually a few revert.
